I am exploring my new Note 10.3 and cannot find a way to control contrast at the device level - for example on the home screen or in Android settings.
Is there a system-wide setting for this, or is contrast only controllable in Neoreader?

Yes, a system wide contrast control is needed!
you can change the contrast of individual apps by clicking on them and optimise settings
but, will only work for that specific app, and will only work if you open it directly from the launcher, if the app is called by another it wont work
example:
open directly from the app drawer chrome with enhanced contrast, will work
open a link in chrome by clicking in gmail application, will not work for the chrome contrast.
